1. Place 50 ul of the sample and 1 drop of each control into separate circles on the card test. 2. Shake the latex vial gently before using. Add 1 drop of ASO Latex reagent to each circle next to be sample tested. 3. Mix with a disposable stirrer and spread over the entire area enclosed by the ring. Use a new stirrer for each sample. 4. Rotate cards at 100 r.p.m. for 2 minutes (see note 2.)
INTERPRETATION OF RESULTS
Examine macroscopically for the presence or absence of clumps within a minute after removing the card from the rotator.
The presence of a visible agglutination indicates a concentration of anti-streptolysin of 200 ml IU/mL. Positive sera may be diluted. Make serial two-fold dilutions in 9g/L saline. The serum titer is defined as the highest dilution showing a positive result.
The approximate ASO level (IU/mL) present in the sample may be obtained by multiplying the titer by the limit of sensivity (200 IU/mL). ASO titers of 200 IU/mL are found in 95% of the normal adult population. Up to 300 IU/mL, are higher values observed among school age children.
