APPLICATION
For screening of syphilis. |

1. Place 50ul of plasma or serum into a ringed slide or onto a circle on the test card using a micropipette. Use a clean pipette tip for each sample. 2. Shake the dispensing bottle assembly, invert and gently tap the needle to remove air bubbles. 3. Position the needle perpendicular to the test card and allow one drop of antigen to fall next to the test specimen. 4. Mix the sample and antigen together 5. Rotate the test card at 100 r.p.m for 8 minutes on a mechanical rotator.
INTERPRETATION OF RESULTS
At the end of 8 minutes of rotation, inspect the test card macroscopically, in good light. Reactive display characteristic agglutination ranging from light (Week-reactive) to intense (Reactive). Very weak reactive results are characterized by small agglutinate around the periphery of the test area. Negatives do not exhibit this reaction and display a macroscopically smooth and even appearance.
1. Using a micro pipette dispense 50ul of physiological saline onto separate rings of a slide or circles of a test card. 2. Using a micro pipette dispense 50ul of the sample (serum or plasma) to be titrated into the saline of the first circle. 3. Using the same pipette mix the saline and sample and transfer 50ul to the saline in the 2nd ring or circle. 4. Perform serial dilutions in the same manner until the last circle and discard 50ul from the final circle. 5. Proceed as in the qualitative test. Circle 1 2 3 4 5 Dilution 1:2 1:4 1:8 1:16 1:32 6. Using a mixer spread the diluted sample across the entire surface of each circle starting at the highest dilution (circle number 5). Proceed to circles number 4,3,2 and 1 in a similar fashion 7. Proceed as in the qualitative test.
